產品介紹 Involved in cytoskeletal rearrangements required for phagocytosis of apoptotic cells and cell motility. Functions as a guanine nucleotide exchange factor (GEF), which activates Rac Rho small GTPases by exchanging bound GDP for free GTP. Its GEF activity may be enhanced by ELMO1.
Tissue specificity: Highly expressed in placenta, lung, kidney, pancreas and ovary. Expressed at intermediate level in thymus, testes and colon.

????乳腺癌是一種常常發生轉移的癌癥。將近90%的乳腺癌患者都因為癌癥擴散而死,因此,理解這一危險過程的細胞和分子基礎機制是十分重要的。
????近日,加拿大蒙特利爾臨床研究所(Institut de recherches cliniques de Montreal, IRCM)的科學家在乳腺癌研究方面取得了一個重要的發現,他們確定了,一種名為DOCK1的蛋白質可以作為潛在的靶標用于減少乳腺癌患者的癌轉移。相關研究結果刊登在了近期出版的《PNAS》上。
????研究人員在乳腺癌治療方面盡管已經取得了重要的進展,但是對于癌癥擴散的機制卻知之甚少。科學家正在試圖確定能夠調節癌轉移過程的蛋白質,因為它們或許可以作為新的藥物靶標,并與現有的方法相結合促進治療的效果。
????在這項研究中,2個主要的癌癥亞型——HER-2+型和Basal-like型,很容易轉移和復發,并最終導致患者的低生存率。該調查以HER-2+(人類表皮生長因子受體2)型為研究對象, 這代表了大約25%的乳腺癌病例。 HER2陽性腫瘤比其它類型的腫瘤更傾向于快速的發展和擴散。
????研究人員解釋到,通過研究HER2+乳腺癌小鼠模型,確定了DOCK1蛋白質是癌轉移的一個重要的調控因子。當去除了小鼠中的這一蛋白質后,乳腺癌的肺轉移大大減少了。此外,研究發現DOCK1蛋白質有助于腫瘤的生長。
????為了證明DOCK1的表達與乳腺癌預后之間的關系,研究人員對包含患者基因信息的幾個數據庫進行了分析。結果發現,在HER-2+型或Basal-like型乳腺癌中,高水平的DOCK1與較差的預后以及疾病的復發有關。
